چکیده

Continuous 14C 0 2 Evolution, Glycolytic Compartment, Pyruvate Metabolism, Synchronous, Yeast The different metabolic behaviour of endogenously produced [1-14C] pyruvate derived from [3,4-14C]glucose and of exogenously added [1-14C]pyruvate were studied with synchronous yeast cell populations, under conditions which differentially influenced the activities of pyruvate decarboxylase in the cytoplasm and pyruvate dehydrogenase in the mitochondria. Endogenously produced [1-14C]pyruvate is decarboxylated almost exclusively by PDC under anaerobic condi tions, in contrast to added [1-14C]pyruvate which is decarboxylated under aerobic conditions by the action of PDH mainly. Whereas 14C 0 2 evolution from exogenous [l-14C]pyruvate can be diluted proportionally by addition of pyruvate, this is not the case for 14C 0 2 evolution from endogenous [1-14C]pyruvate. It is suggested that glycolysis or at least some constituents of it might be arranged in such a manner that the resulting complex behaves as a functional compartment, meaning that it is almost inaccessible to exogenously added pyruvate.

O24: Functional Role of the K2P Potassium Channel TASK-3 in Glioma

TASK-3, a two-pore-domain (K2P) potassium channel, has been implicated as important regulator for the effector function and proliferation of T-cells. Interestingly, TASK-3 has also a functional impact on tumor cells. Therefore, we sought to investigate whether TASK3 modulation might have a therapeutic potential for malignant gliomas by a variety of phenotypical and functional in vitro assays mi...

In Vivo Effects of Calcitriol on Phenotypic and Functional Properties of Dendritic Cells

Background: Dendritic cells (DCs) play a central role in the initiation and expansion of T cell mediated immune responses with potential immunotherapy application. The compounds which have the ability to induce immunomodulatory effects on DCs may be employed for the treatment of immunopathologic conditions such as autoimmune diseases. Objective: The aim of this study was to investigate the in v...

Dynamic regulation of yeast glycolytic oscillations by mitochondrial functions.

The control exerted in vivo by mitochondrial functions on the dynamics of glycolysis was investigated in starved yeast cells that were metabolizing glucose semianaerobically. Glycolytic oscillations were triggered after a pulse of glucose by inhibition of mitochondrial respiration with KCN, myxothiazol and antimycin A or in mutants in the bc1 complex (ubiquinol:cytochrome c reductase) that were...

Antagonistic Activity of Fructoplane Yeast Against Ulocladium Rot of Papaya

Debaryomyces hanseniZopf isolated from the fructoplane of apples were found to be effective as biocontrol agent against rot of papaya caused by Ulocladium. chartarum(Pr.) Simm. The ability of D. hansenii to prevent infection of U. chartarum was lost when the antagonist cells were killed by autoclaving. Cell free culture filtrates of antagonist were unable to prevent disease incidence. Efficacy ...

Respiratory mutant and liquid holding recovery inhibition in yeast cells

Background: Cell ability to recover from radiation damage is of great relevance in cancer treatment. It is often believed that the inhibition of cell ability to the liquid holding recovery (LHR) may be an indicator of the overall suppression of cell ability to recover from potentially lethal radiation damage. However, the literature contains no experimental evidence whether the LHR inhibition m...
